CHROMATOGRAPHY: Chromatography is based on the Greek wordchroma, for colour. This technique is extensively used to separate mixtures into their components, purify compounds and also to test the purity of compounds. This is classified into differentcategories. Two of these are: 1 Adsorption chromatography, and 2 Partition chromatography. 1 Adsorption Chromatography: The main principle is that, different compounds are adsorbed on anadsorbent to different degrees. Commonlyused adsorbents are silica gel and alumina.When a mobile phase is allowed to moveover a stationary phase adsorbent,the components of the mixture move byvarying distances over the stationaryphase. The two main types ofchromatographic techniques based on theprinciple of differential adsorption are: 1 Column chromatography, and 2 Thin layer chromatography.Column Chromatography: Columnchromatography involves separation of amixture over a column of adsorbentstationary phase packed in a glass tube. Thecolumn is fitted with a stopcock at its lower end. The mixture adsorbed on adsorbent is placed on the top of theadsorbent column packed in a glass tube. Anappropriate eluant which is a liquid or amixture of liquids is allowed to flow down thecolumn slowly. Depending upon the degreeto which the compounds are adsorbed,complete separation takes place. The mostreadily adsorbed substances are retained nearthe top and others come down to variousdistances in the column.
