DNA isolation from fish.
This is a practical video about DNA isolation from fish performed by bio-technologists. While sampling according to the requirement weigh the sample, here we have taken 2 gm of fish fin tissue sample. You can store the sample by adding 95% ethanol or can use fresh sample as like ours. Add lysis buffer 500µl (Tris HCL, EDTA, SDS) and homogenize until fine crushed. Add proteinase k 2µl and vortex the sample. Incubate the sample overnight. Add equal amount of Phenol chloroform isoamyl-alcohol (25:24:1) and mix it thoroughly by inversion for 5mins. The sample was centrifuged at 10000rpm for 10 min at 27 °C, and the supernatant was placed into a new, sterile mirco-centrifuge tube. Repeat the PCI step and centrifuge it again at 10,000rpm for 10 min. Transfer the supernatant into fresh tube and add absolute ethanol, centrifuge 10000rpm for 10 min at 4 °C. Discard the supernatant and air dry the pellet. Add 30µl of Nuclease free water to dissolve the pellet and store at -20°C. Run the samples on 0.8% agarose gel for electrophoresis for 45min at 80 volts. After electrophoresis you can visualize the gel under gel documentation unit. Amplicons/bands of DNA observed, confirms the presence of isolated DNA.
