Meso Gaurd requires at least Chakra to be level 3. Meso Explosion can be any level it wants to be, depending on the character build. Here's my build at the time the video was made.
Level 70 - Assaulter
Level 71 - Meso Explosion, Chakra, Band of Thieves
Level 72 - Chakra, Chakra, Meso Guard
Level 73 - Assaulter, Assaulter, Assaulter
Level 74 - Assaulter, Assaulter, Assaulter
As you can see, I got all the fun skills first, then I proceeded to max out Assaulter.
