Shotgun DNA sequencing is not a specific DNA sequencing method like any other method of DNA sequencing. Its a procedure for complete genome sequencing. Here, the whole genome is fragmented into smaller segments (reads) randomly and each small segment of DNA is sequenced separately using any DNA sequencing method like Sanger’s dideoxy DNA sequencing. This process is repeated 3 to 4 times using different chemicals or enzymes to fragment DNA. Later, by using the overlapped regions of these fragments, a computer program aligns these sequences properly so that to sequence the whole genome or chromosome.
