The wound healing or scratch assay is a standard technique for probing collective cell migration in two dimensions. A monolayer of cells are scratched with a pipette tip. The migration of cells into the gap is imaged over several hours using a microscope. The primary information derived is the rate of gap closure, a measure of the speed of the collective motion of the cells.
