Pyrosequencing
Hello Dear learners In this lecture we will discuss about Pyrosequencing. It is a DNA sequencing method that was widely used in research and clinical applications. It has been largely replaced by newer sequencing technologies like Illumina's sequencing-by-synthesis (SBS) technology. However, it remains an important historical method in the field of genomics. Pyrosequencing involves the real-time detection of nucleotide incorporation during DNA synthesis. It is a sequencing-by-synthesis method that does not require labeled nucleotides. Instead, it uses a series of enzymatic reactions to measure the release of inorganic pyrophosphate (PPi) when a nucleotide is added to a growing DNA strand. The release of PPi leads to the production of light, which is quantified to determine the sequence of the DNA template.
